I dunno if I can help you much here. Most of my work is completely digital. It’s been a while since I did traditional art.
However when I do I usually do a hi-res scan and desaturate it. I then adjust the contrast/curves, making sure the lineart is fairly crisp and high contrast and the background is a pure white.
I then open the channels panel and press ‘command’ and click the thumbnail on the RGB layer. This should load a selection that looks something like this.

I then hit delete, which should now remove the white. You can now go back to your normal layers panel and lock the alpha, then colour over your lines in black or whatever colour you want your lineart to be!
They usually end up a little thicker/heavier than before so you may ned to go all the way back and adjust the contrast more. Once you’re happy you can then colour below that layer without needing to apply a blending mode.
